Es ist kein Geheimnis mehr, dass die Erdölreserven begrenzt sind und die Förderung immer problematischer wird, bevor sie Mitte oder Ende des 21. Jahrhunderts ganz zur Neige gehen. Der Aufwand steigt und damit zugleich die Kosten, außerdem ist die Förderung und der Transport über die Meere in Tankern mit bis zu 300.000 t Öl oder über weite Strecken in Pipelines mit hohen Risiken verbunden.
Kürzlich zeigte uns das die Ölkatastrophe im Golf von Mexico. Seit dem 22. April 2010 sind schätzungsweise bis zu 575.000 t Rohöl aus einem Bohrloch ausgeströmt. Erst im Dezember 2009 wurde bekannt, dass bei der Erdöl- und Erdgasförderung jährlich Millionen Tonnen radioaktiv verseuchter Rückstände anfallen, für dessen Entsorgung größtenteils der Nachweis fehlt.
Pflanzen, und vor allem crops, als Rohstoffquelle sind nicht unumstritten, das treibe die Weltmarktpreise der Lebensmittel in die Höhe, mahnen Kritiker. Doch angesichts der wirtschaftlichen Schäden, den Förderung und Transport von Rohöl sowie Klimawandel verursachen, wird niemand mehr bestreiten, dass Pflanzen nicht mehr nur eine ökologische sondern auch eine ökonomische Alternative darstellen.
Einen weiteren Schritt in Richtung Wirtschaftlichkeit geht das Konzept der Bioraffinerie. Die Logik dahinter ist so einfach wie bestechend: Hier werden die Pflanzen nicht nur für einen Zweck genutzt. Die unterschiedlichen Bestandteile werden der Reihe nach für verschiedene Zwecke verwendet; die Pflanze wird so komplett verwertet. Der analoge Gedanke liegt seit Jahrzehnten den Ölraffinerien zugrunde.
From Biomass to Feedstock
Biomasse ist aus verschiedenen organischen Verbindungen in sehr unterschiedlichen Anteilen zusammengesetzt, hauptsächlich Lipide, Saccharide und Proteine. Daneben finden sich zahlreiche weitere Verbindungen, die aber meist in geringeren Anteilen vorkommen, wie z. B. die Sekundärmetabolite. Mit Hilfe der Bioraffinerie und biotechnologischen Verfahren werden daraus verschiedene Produkte und höherwertigere Verbindungen: Nahrungs- und Futtermittel, Grund- und Feinchemikalien für die chemische Industrie oder Energie in Form von Kraftstoffen, Strom oder Wärme aber auch Bioplastik.
Um die Rohstoffe aufzubereiten, bestimmte Fraktionen zu isolieren und mit chemischen und biotechnologischen Verfahren weitere Verbindungen abzuleiten, wird die Biomasse zuerst zerkleinert, getrennt und isoliert. Mit diesem Verfahren lassen sich bereits in der Biomasse vorhandene Stoffe und Verbindungen gewinnen. Durch chemische Veränderung lässt sich das Produktspektrum erweitern.
Neben der so genannten Pyrolyse, der thermischen Spaltung durch hohe Temperaturen, kann die Biomasse durch Fermentation umgewandelt werden, also mit Hilfe von Bakterien-, Pilz- oder Zellkulturen oder Enzymen. Die Umsetzung und Beschleunigung oder Lenkung chemischer Reaktionen, in der Enzyme als Katalysatoren dienen, wird als Biokatalyse bezeichnet. Die Enzyme werden entweder isoliert oder in der lebenden Zelle zur Katalyse von chemischen Reaktionen verwendet.
Die Vorteile der Biokatalyse für die organische Synthese liegen vor allem in der Selektivität, die eine Voraussetzung für eine hohe Ausbeute ist. Enzyme reagieren oft unter milden Bedingungen, so dass bei gegebenen Reaktionsbedingungen nur eine funktionelle Gruppe reagiert. Dadurch sind die Produkte biokatalytischer Prozesse oft reiner und die Abtrennung von Nebenprodukten ist unproblematischer. Durch die Anpassung der Enzyme an die spezifischen Prozesse der Biokatalyse bietet das Verfahren noch ein enormes Potenzial, die Ausbeuten deutlich zu erhöhen.
Optimierte Prozesse eine verbesserte Wirtschaftlichkeit
Bei der Erzeugung von Kraftstoffen der ersten Generation wurde mit Öl, Zucker oder Stärke nur ein kleiner Teil der Pflanze genutzt. Mit Hilfe der Bioraffinerie entsteht weniger Konkurrenz zur Ernährung. Bei Kraftstoffen der zweiten Generation wird nahezu die vollständige Pflanze verwendet, einschließlich der schwer zugänglichen Cellulose. Kraftstoffe der zweiten Generation sind energie- und ressourceneffizienter, erfordern jedoch einen höheren technischen Aufwand und nur optimale Prozesse ermöglichen eine konkurrenzfähige Produktion.
Bei der Herstellung von Ethanol auf Zellulosebasis sind die Enzyme eines der Haupthindernisse. Die Vorbehandlung war bisher zu teuer und die Kosten der zuckerbildenden Enzyme musste verringert werden. Die Verzuckerungs-Effizienz von Hemizellulose und Zellulosezucker musste verbessert werden sowie die effiziente Nutzung von sowohl Glucose als auch Xylose. Schließlich musste die Wirtschaftlichkeit der Neben- und Koppelproduktströme optimiert und der Maßstab erfolgreich vergrößert werden.
Bei den biobasierte Kunststoffen wird mit Hilfe von Fermentation ein völlig neuer Werkstoff hergestellt, mit sehr exakt einstellbaren Eigenschaften. Als Ausgangsstoffe für Biokunststoffe dienen aktuell vor allem Stärke und Cellulose als Biopolymere von Zuckern.
Die Polymilchsäure (Polylactid, PLA) entsteht durch Polymerisation von Milchsäure, die wiederum ein Produkt der Fermentation aus Zucker und Stärke durch Milchsäurebakterien ist. Das durchsichtige Material gleicht herkömmlichen thermoplastischen Massenkunststoffen nicht nur in seinen Eigenschaften, sondern lässt sich auch auf den vorhandenen Anlagen ohne weiteres verarbeiten. Das Biopolymer Polyhydroxybuttersäure (PHB) ist ein fermentativ herstellbarer Polyester mit Eigenschaften ähnlich denen des petrochemisch erzeugten Kunststoffs Polypropylen. Es kann auf Basis von Zucker und Stärke hergestellt werden, die Synthese ist jedoch auch aus anderen Nährstoffen wie Glycerin und Palmöl möglich.
Die organischen Verbindungen der Biomasse können aber auch in der Bioraffinerie zu Ethanol fermentiert werden. Durch Cracken, eine Standardprozedur in der Petrochemie, wird daraus Ethen und durch Polymerisation Polyethylen (PE). Wie auch bei den Biokraftstoffen müssen hier für eine verbesserte Wirtschaftlichkeit die bestmöglichen Prozessbedingungen und die optimalen Organismen für die Fermentation gefunden werden.
University of Iowa
Seit mehr als 20 Jahren forschen Wissenschaftler am „Center for Biocatalysis and Bioprocessing“ (CBB) an der University of Iowa auf den Gebieten der Bioverfahrenstechnik und Biokatalyse. The principal areas of study include biocatalyst fundamental properties, bioremediation, bioprocessing, new biocatalyst discovery, novel biocatalyst applications, biosensing technology, and reactive agent development. Seit April 2007 setzt das Institut für die Forschung die parallelen DASGIP Bioreaktoren ein, um damit die Enzyme für die Biokatalyse zu optimieren.

The CBB laboratory brings together the University of Iowa core fermentation facility, laboratory space for conducting industry/university related research and a first-rate facility for delivering short courses and training associated with the CBB,” said Professor Mani Subramanian, Director of the CBB.

Da kein anderer Staat der USA mehr Mais und Soja und infolgedessen Bioethanol und Biodiesel produziert als das landwirtschaftlich geprägte Iowa, liegt hier auch ein Schwerpunkt der Forschung an der CBB. Es zeigte sich, dass sich die Körner, Bohnen sowie Pflanzenreste bei der Herstellung von Grund- und Feinchemikalien hervorragend ergänzen. An der University of Iowa wurden so vor allem neue aromatische Verbindungen aus nachwachsenden Rohstoffen entwickelt mit neuen chemischen Eigenschaften und verbesserter Leistungsfähigkeit.

Technologie
Moderne Technologie ermöglicht Wissenschaftlern die Entwicklung von Biokunststoffen und Biokraftstoffen, die nicht nur eine ökologischer sondern auch zunehmend eine wirtschaftliche Alternative darstellen. Mit den DASGIP Parallelen Bioreaktorsystemen erhalten die Entwickler ein Werkzeug, das an die spezifischen Anforderungen der Prozess- und Enzymoptimierung ausgerichtet ist. So erlaubt das System die Temperaturregelung bis 99°C – eine Temperatur, die für die Spaltung der Zellulose zu Stärke und Zucker notwendig ist.
Anaerobe Fermentation sowie spezifische Mess- und Kontroll-Anforderungen werden erfüllt: die wichtigsten Parameter wie pH-Wert und Redox-Potenzial, Gasversorgung wie Methan werden kontinuierlich überwacht und die Automatisierung der komplexen Abläufe ist möglich. Das ermöglicht das Screening nach kosteneffizienten Enzymen für unterschiedlichste Saccharide unter Berücksichtigung bereits bestehender Fertigungsprozesse. Wissenschaftler können die Leistung von verschiedenen Mikroorganismen unter gleichen Bedingungen, oder gleichen Mikroorganismen unter verschiedenen Bedingungen verglichen. Die hohe Informationsdichte und den parallelen Betrieb von bis zu 16 Reaktoren optimiert das Verfahren und beschleunigt Auswahl der Stämme sowie der besten Prozessparameter. Ein Up-Scale vom Labor- auf den Industriemaßstab ist in einem Schritt und ohne Aufwand durchführbar.
Die Kombination aus der Vielzahl an zentral überwachten und geregelten Parametern mit der Vielzahl an parallel gesteuerten Reaktoren macht das DASGIP System darüber hinaus zum hervorragenden Instrument bei der Umsetzung von Methoden der statistischen Versuchplanung (DoE).

It is no longer a secret that the world is sitting on limited oil reserves and that before supplies completely dwindle by the middle or late 21st century, extraction will become increasingly problematic. The effort, and thus the costs, are rising. In addition, there are major risks associated with extracting and transporting up to 300,000 tons of crude oil with ocean tankers or across long-distance pipelines.
This was recently illustrated by the oil catastrophe in the Gulf of Mexico. Estimates are that up to 575,000 barrels of crude oil have spewed out of the Deep Horizon wellhead since April 22, 2010. The fact that oil and gas exploration leaves behind millions of tons of radioactive residue each year with little evidence of its disposal, first became known in December 2009.
The use of plants, and especially crops, as a source of raw materials is not undisputed. Critics maintain that it drives up global food prices. However, in light of the economic damage that results from the extraction and transportation of crude oil and climate change, any doubts that plants represent not only a viable environmental, but economical alternative, have vanished.
Another step in the direction of economic viability is the concept of the biorefinery. The underlying logic is as simple as it is captivating. The plants are fully recycled, with the different parts of the plant being utilized for different purposes. Oil refineries have used this same thought process for decades.
From biomass to feedstock
Biomass is made up of various organic compounds in very different ratios, primarily lipids, saccharides and proteins. Numerous other compounds such as secondary metabolites can also be found, but mainly in smaller ratios. A variety of products and valuable compounds are derived through biorefinery and biotechnology processes. Among them are food and feed stuffs, basic and fine chemicals, and energy in the form of fuels, power, heat and even bioplastics.
To prepare the raw materials, isolate specific fractions and derive additional compounds using chemical and biotechnology processes, the biomass first has to be crushed, separated and isolated. This process yields the materials and compounds already contained in the biomass. The product spectrum can be expanded by means of chemical change.
In addition to so-called pyrolysis, which is thermal decomposition by means of high temperatures, the biomass can be transformed via fermentation; in other words with the help of bacteria, fungi and cell cultures or enzymes. The initiation and acceleration or modification of chemical reactions in which enzymes serve as catalysts is referred to as biocatalysis. The enzymes are either isolated or utilized in the living cells for the catalysis of chemical reactions.
The major advantage of using biocatalysis for organic synthesis lies in the selectivity, which is a prerequisite for a high yield. Enzymes frequently react under mild conditions, so that only one functional group responds under given reaction conditions. Products derived from biocatalytic processes are therefore often more pure and the separation of the byproducts unproblematic. By adapting the enzymes to the specific biocatalytic processes, this approach improves the already-enormous potential to significantly increase the yield.
Optimized processes yield improved cost-efficiencies
The production of first-generation fuels involved using only a small part of the plant, together with oil, sugar or starch. The use of biorefineries lessens the impact of diverting these food stuffs from the human food chain. To produce second-generation fuels, nearly the entire plant is used, including the cellulose, which is difficult to break down. While these fuels are more energy and resource efficient, they require a greater degree of technical complexity. Implementing optimized processes is the only way to ensure production that can effectively compete with conventional methods.
Enzymes are one of the main obstacles to producing ethanol with cellulose. To date, pre-processing has been too expensive and has required finding ways to reduce to the costs of the saccharifying enzymes. The saccharification efficiency of hemi-cellulose and cellulose sugar, as well as the efficient utilization of glucose and xylose, had to be improved. Finally, the cost effectiveness of the coproduct and byproduct energy had to be optimized and successfully scaled up.
Bioplastics utilize fermentation to produce a completely new material with properties that can be precisely defined. Today, polysaccharides such as starch and cellulose are the most common source of materials for producing bioplastics.
Polylactic acid (PLA) is created through the polymerization of lactic acid, which in turn is derived by fermenting sugar cane and corn starch with lactic acid bacteria. This transparent material not only has properties similar to conventional thermoplastic mass polymers, it can be processed without modifying existing plant equipment. Polyhydroxybutyrate (PHB) is a fermentation-based biopolymer with properties similar to polypropylene, a petrochemical plastic. Although PHB can be produced with sugar and starch, the synthesis requires other nutritive substances such as glycerin and palm oil.
Biomass organic compounds can also be fermented into ethanol in biorefineries. Ethene is derived through steam cracking, a standard petrochemical process, while polyethylene is produced by means of polymerization. As with biofuels, biomass-based ethanol requires optimal process conditions and fermentation organisms to make it economically viable.

Scientists at the Center for Biocatalysis and Bioprocessing (CBB) at the University of Iowa have engaged in the research of bioprocessing technologies for more than 20 years. The principal areas of study include biocatalyst fundamental properties, bioremediation, bioprocessing, new biocatalyst discovery, novel biocatalyst applications, biosensing technology and reactive agent development. The center has relied on parallel bioreactors from DASGIP since April 2007 to optimize enzymes used in biocatalysis processes.

DASGIP System for biomass and protein optimization and reaction engineering
Iowa and its heavily agriculture-based economy produces more corn and soy beans, and as a result more bioethanol and biodiesel, than any other state in the country, thus providing the CBB an additional research focus. Research has shown that seeds, beans and the remains of plants highly complement one another in the manufacture of basic and fine chemicals. With this in mind, the University of Iowa developed new, renewables-based aromatic compounds that exhibit new chemical properties and improved efficiency.

Technology
Modern technology enables scientists to develop bioplastics and biofuels that represent not only an environmentally-sustainable, but an increasingly cost-effective alternative. DASGIP parallel bioreactor systems provide developers a tool that is designed specifically to meet the needs of process and enzyme optimization. As an example, system temperatures can be regulated up to 99°C, the temperature at which cellulose can be broken down into starch and sugar.
The system provides an optimum environment for anaerobic fermentation, including the specific measurement and control requirements. Key parameters such as pH, redox potential and gas levels such as methane are continually monitored. All of the complex processes can be completely automated. This makes it possible to screen for the widest variety of cost-effective saccharide enzymes in existing manufacturing processes. Scientists can compare the efficiency of different microorganisms under the same conditions or vice versa. The high information density and parallel operation of up to 16 reactors optimizes the process and speeds up the selection of the cells and the best process parameters. With minimal effort, laboratory systems can be scaled up to industrial size in one step.
Moreover, the wide selection of centrally-monitored and regulated parameters combined with the large number of parallel reactors makes the DASGIP system an excellent choice for carrying out statistical test planning (DoE).
